چکیده

by oncogenic viruses [simian virus 40 (SV40) or jx)lyoma virus or sequential infection with both viruses], their susceptibility to changes caused by L-fucose was greatly reduced. Thus, virustransformed cells grown in the presence of L-fucose showed much less of a change in the morphology of individual cells, their pattern of association, their rate of growth, and incorporation of protein and RNA precursors as compared to ordinary 3T3 cells. In the different cell lines tested, there was a general paral lelism between degree of susceptibility to alteration by L-fucose and degree of contact inhibition. The mechanism by which Lfucose alters ordinary 3T3 cells, and the reasons for the difference in virus-transformed lines are unknown. However, the results raise the possibilities that L-fucose alters ordinary 3T3 cells by combining with complementary sites of these cells and that interactions between natural cellular sugar constituents and complementary sites play a role in mediating cell contact in hibition.

The Validated Embryionic Stem Cell Test to Predict Embryotoxicityinvitro

Backgrounds: A straight-forward way to identify whether a drug or environmental chemical can be harmful to unborn baby is to evaluate its effect on laboratory animals. All invivo methods need large number of animal and are therefore time consuming and expensive. However, the thousands of chemicals in need of testing, to reduce the spending of live animals, an assortment of in vitro assays has ...

Enhanced Na+-K+-activated adenosine triphosphatase activity in transformed fibroblasts.

Differences in membrane fluidity and structure in contact-inhibited and transformed cells.

Studies on contact-inhibited mouse embryo fibroblast 3T3 cells and 3T3 cells transformed by oncogenic RNA and DNA viruses and by a chemical carcinogen have demonstrated differences in plasma membrane architecture. Spin-label and freeze-fracture ultrastructural studies have shown that contact-inhibited cells have ordered membrane lipids and aggregated intramembranous particles, whereas transform...

Mobility of normal and virus-transformed cells in cellular aggregates

The mobility of embryonic chick cells and cells of four established cell lines was examined in cellular aggregates. This was done by preparing aggregates of unlabeled cells and allowing cells of the same type, but prelabeled with [3H]thymidine, to adhere to the surface of the aggregates. After 2-1/2 days in agitated liquid culture the positions of the labeled cells within the aggregates were de...

Effects of nitrosocarbaryl on BALB/3T3 cells.

Carbaryl(N-methyl-1-naphthylcarbamate) and its nitrosated product, N-nitrosocarbaryl, were tested for their effects of BALB/3T3 (clone A31) cells in culture. Nitrosocarbaryl, but not carbaryl, caused transformation of the BALB/3T3 fibroblasts, but neither chemical induced the complete expression of endogenous murine leukemia virus. Transformed cells differed from the parental control cells by l...
